> ATERA – Reconstructed Human Gingival Epithelium

The ATERA – Reconstructed Human Gingival Epithelium (RHG) consists of normal human-derived gingival mucosal cells cultured for 5 days at the air-liquid interface, resulting in a multilayered mucosal equivalent in vitro that is histological similar to the human gingival epithelium. The ATERA – RHG is characterized by a high metabolic activity and can be employed for safety and efficacy testing. To ensure global availability, models are transported on an agarose based solidified medium together with ATERA – growth and maintenance medium.

Culture

The ATERA – RHG is generated from normal human-derived gingival mucosal cells that are seeded on a synthetic carrier membrane. To mimic the in vivo situation, the tissues are reconstructed at the air-liquid interface. Due to these conditions and a chemically defined medium, a natural differentiation is triggered and a multilayered gingival mucosa is formed. In the standard configuration e.g. to test irritative effects of dental products, the models are cultured under these conditions for 5 days. However, depending on the application, the ATERA – RHG can also be used after different periods.

The ATERA – RHG recapitulates the histological architecture of human gingival mucosa. Besides several layers of undifferentiated cells in the lower part of the model, a layer of differentiated flattened cells is detectable in the outer layer of the model. However, comparable to the in vivo situation, no stratum corneum is detectable.

The production of the ATERA – RHG is performed at Sterlab France. To ensure reproducible results for our customers, high quality standards are employed. Models are produced in a biosafety level 2 clean room that is accredited according to ISO 9001 V2008. All used cell batches are tested before use for Hepatitis B, Hepatitis C, HIV, and Mycoplasma. Furthermore, only model batches are provided that comply with our internal quality criteria listed below:

The ATERA – RHV is generated from of the human epithelial cell line A431. Following an in vitro expansion step, cells are seeded on a synthetic carrier membrane and the tissues are reconstructed at the air-liquid interface.
